com.sun.tools.javac.Main is not on the classpath.

Perhaps JAVA_HOME does not point to the JDK.

It is currently set to "C:\Program Files\Java\jre6"

What about setting the JAVA_HOME to your jdk?

 

Eclipse>Window>Preferences>Java>Installed JREs>Add...>Next>Directory...>Computer>C:\>Program Files>Java>jdk1.x.x_x>OK>Tick your jdk instead of jre.
